Dapagliflozin: FDA Wants More Data for Approval

A drug treatment for Cyctic Fibrosis was approved by FDA. The drug was developed after a gene responsible for causing Cycstic Fibrosis was found. The identification of gene has been acomplished after almost 22 years.

Vertex is producer of the drug named as Kalydeco. Cystic fibrosis is caused because of defective gene responsible for decoding a protein known as Cystic Fibrosis Transmembrane Regulator. It is found that the gene is behind 4% cases of cystic fibrosis.

Approval of Kalydeco is believed to be of great significance as all other available treatments in market are only capable to treat symptoms of disease. A press note has been published by USA FDA regarding the approval.

The treatment is approved for patients of age above six and with confirmed G551D mutation. Vertex is quite please as approval came earlier than expected.

Considering rareness of the disease, producer company is contemplating to initiate programs to make the drug available at affordable cost or may be for free. Current estimation of drug cost is $294,000 per year which is near to the cost of other treatment for same disease.

Kalydeco is expected to generate millions of dollars in US market alone. There are 200,000 patients of Cystic Fibrosis in USA.

Recent amendment in FDA approval policy is the reason that the drug was approved with priority in review.
Clinical trial included 213 patients with G551D defect. The pill was administered twice a day in all participants. During trial Kalydeco showed some side effects such as stomachache, headache, infection in respiratory system and diarrhea.

Cystic Fibrosis is serious disease as it decreases life of patients because of thick mucus accumulated in digest tract and lungs. Patient suffering from CF lives upto 36-38 years.

Fast Pain Reliever Method Under Trial

Before any surgery patients are given anesthesia to numb the portion of the body. That is to reduce the pain while performing operation. But for treatments, such as root canal of teeth, cosmetic surgeries, short acting anesthesia is used on patient. Short term anesthesia has couple of shortcomings such as effect last for few hours, impaired eye sight and unable to eat or talk.

A new development has been made by experts to subdue problems caused by short acting anesthetic. Scientists have developed a new molecule which can be used as fast acting pain relievers. The molecule, as demonstrated at Nature, works by getting activated when shed a beam of light on it. Activated molecule silences pain causing neurons. The molecule is then deactivated by light to terminate its effect. This new instant pain reliever is believed to be significant progress and likely to be most favorite pain reliever in general clinics.

General anesthetics bring numbness by blocking electrical signals generated by ending of nerve cells. The endings of nerve cells are medically known as Nociceptors. Nociceptors generate signals when cold, heat or mechanical pressure exceeds threshold limit. These generated signals are due to reach central nervous system which, after following numerous chain reactions, eventually results in feeling of pain.

Most commonly used anesthetic is Xylocaine. It works by blocking nociceptors from generating signals of pain. There are many other local anaesthetics also and all of them work in somewhat similar way. Problem with these current anesthetics is regardless of specific nociceptors they act on all the neurons. The new molecule based pain reliever method is selective in nature. With that being said, the method acts only on desired area of the body.

The development of new molecule known as Ammonium-azobenzene-quaternary ammonium is a result of combined efforts by scientists of California Uni and Ludwig-Maximilians Munich. Dr Richard Kramerof California and Dr Dirk Trauner are lead researcher of their respective group. The molecule changes its shape when exposed to green light and returns to its original shape when exposed to ultraviolet beam of light. The change in shape occurs within hundreds of part of a millisecond. This makes the molecule super fast acting pain reliever.
The trial of the new method has been successful.

Merck's New Anti-Platelet Drug Produce Mixed Result

A new antiplatelet drug has shown significant ability to prevent blood clots. However, the drug also caused abnormal bleeding in clinical trial. Merck & Co is testing the drug in a trial which is believed to be largest drug trial conducted for any heart medicine so far.

The manufacturer company is seeking approval for Vorapaxar drug since last year. But the approval is denied because of bleeding caused by the drug in patients with history of stroke. Submitted data of the study included 26,449 participants.

Merck has been asked to stop the trial on patients who had stroke in recent past and, allowed to continue trial on heart attack patients who are in stable condition at present. Vorapaxar is currently being tested on patients who have recently been diagnosed with clogged leg arteries.

Latest trial data published by drug maker last month showed significant reduction in forming of blood clots, while on the other hand, it caused excessive bleeding. Experts are inconclusive whether the company would apply for approval. A trial named TRACER for same drug had failed too. It is also not clear whether Merck would conduct another trial on heart attack patients only.

Despite drug maker is quite happy with trial result, experts see very less possibility for approval. Drug usage will be limited to small group even if Vorapaxar succeeds to get approval.

BEST FOR HEART ATTACK PATIENTS

Researchers involved in trial claimed that after 30 months administered Vorapaxar met goal when it's effects were compared with other popular antiplatelet drug such as Plavix and Aspirin. In fact Vorapaxar reduced the risk of stroke, heart attack and cardiovascular diseases by 13% when compared with its other counterparts in market.

In past several decades Vorapaxar is first new antiplatelet drug which provides long time protection for to patients with history of heart attack, says Dr. David Morrow - lead researcher of the trial.

The Food and Drug Administration sanctioned the prescribed drug according the main psychoactive ingredient in arijuana.

Delta 9-THC and Cannabidiol are the two very main elements of Sativex, and got approved by Canada, New Zealand and eight European countries for releasing muscle contraction associated with variety of sclerosis.

Vital and Innovative bond with marijuana was uneasy to represent by FDA for approval. Where as many nations like Washington DC already allowed in use legally prescribed recommendation with doctors.

As the organic substance parallel with prescribed drug, it could raise the burden on government returning its status and potential that, other drug industries will follow GW Pharma. The US Drug Enforcement Administration categorises tub as a warning without medical consequences.

A founder of the company GW Pharma’s Dr. Geoffrey Guy got the permit for the growth and development of prescribed drug as marijuana is yet banned in UK.

Dr Guy bring in the idea at a scientific meetings which brought unreliable proof that tub heals to various sclerosis patients, British government allowed it as a conceivable path for drawing straight line in recreational and medicinal usage, urged Mark Rogerson Industries spokesperson.

New Drug (Jakafi) Approved by the FDA to Treat Myelofibrosis

No more is Myelofibrosis a threatening disorder. After so many years of research and waiting, the researchers have been able to incept a medication that can treat this rare bone disorder known as Myelofibrosis. The FDA (Food And Drug Administration) has never before approved any medication to treat this disorder. Jakafi (Ruxolitinbin) is the first successful medication that is approved by the necessary standard boards to treat the same.

Jakafi manufactured by Incyte Corporation has to be consumed twice a day. This Incyte Corporation has its partnership with Swiss Drugmaker Novartis.

Myelofibrosis is this bone disorder where one has scares instead of the marrow that has to be present. This means the body is unable to produce the necessary blood cells. One is prone to problems like anemia and any sort of other infections in the body. One can also suffer from extrameddulary hematopoiesis. Though there is no particular cause of this severe disorder, it can result because of leukemia or lymphoma.

Richard Pazdur who is the Director of the Office of Hematology and Oncology Products in the FDA Department has been appreciating the process of the invention of this medication where it is based on the process of understanding of disorder and the problems that the sufferer undergoes. generic viagra

This medication was out on a comparative study along with the placebo effect and other sorts of therapy like hydroxyureo. Jakafi however proved to be successful amongst all with patients experiencing 50% better results. Though like every medication, Jakafi can also have its own list of adverse effects including nausea, fatigue and thrombocytopenia.

First FDA approved Anti-venom Medicine

After a relatively small trial FDA has approved anti venom medicine known as Anascorp. Anascrop (an intravenous therapy) is indicated to treat venom of Arizona bark scorpion. The medicine which was available before this approval was not approved by FDA as the treatment was producing severe side effects such as Anaphylaxis (acute multi-system).

Bark scorpion is most venomous one. Its sting can cause paralysys in stung part, severe pain, mouth frothing and vision problem.

Earlier, researchers could not get FDA approval for Anascorp because of lack of clinical trials. There were some obstacles to have participants as exposing people to highly venomous sting was potential risk. But Dr Leslie Boyer knew how important anti-venom was and she searched for hospitals that were ready carry out clinical trial on patients stung by Bark Scorpio. Dr Leslie decided to carry out an open trial.

Initially Dr Leslie found only two hospitals ready to participate. In a year, fifteen kids were successfully treated with Anascorp. Wondering effects were seen with even a small dose of Anascorp. Symptoms of Bark scorpion sting were treated in eight children within 3-4 hours. Patients without Anascrop had to stay longer in ICU before they were declared out of danger.

The trial was started in 2008 and lasted till 2010. 200 children were treated with the anti-venom drug. Trial is longest believed to be the longest running anti-venom trial. An approval based on such unusual trial is also a remarkable one in the history of FDA.

FDA Approves Development of Drug for Wegener's Granulomatosis

(Weston-Based) and Genentech (California) won an approval from U.S. Food and Drug Administration to develop a drug as a part of a new treatment for Wegener's Granulomatosis and Microscopic Polyangiitis diseases. These are very rare and fatal diseases.

The drug Rituxan (FDA approved) is used to treat rheumatoid arthritis, chronic lymphocytic leukemia and non-Hodgkin’s lymphoma. This drug can now also be used in combination with corticosteroids - to treat two other diseases - Wegener’s Granulomatosis and Microscopic Polyangiitis. These diseases impact small blood vessels of the lungs, kidneys, sinuses and other organs. There are many people in U.S. afflicted with these diseases.

Rituxan can be used as an alternate treatment for the CYC chemotherapy treatment for the diseases. The CYC chemotherapy and Rituxan have shown similar results at six months. Based on study and results FDA has approved Rituxan to be used for the same.

The FDA had alerted in 2006 informing that two patients taking the drug (Rituxan) to treat systemic lupus erthematosus were not approved to use the drug. Both the patients had died due to an infection of the central nervous system known as Progressive multifocal leukoencephalopathy. Treating Systemic lupus erythematosus with the drug is not appoved use of the drug.

In 2010, a man sued Biogen and Genentech, as he suffered an infection which paralyzed him. He said that Rituxan had made him more prone to such infections. He was under the medication for thrombotic thromboctopenic purpura, a blood disease that is very rarely found. Rituxan is not an approved drug for the same.

Escalating drug prices, Time to Revise FDA Approval Process?

Few weeks back, Jim Doyle, the reporter of St Louis Post Dispatch came together with a series of compelling accounts on KV Pharmaceuticals. It had got to do with the mystifying economics of the approval by the FDA and with producers of the drug as well. Doyle has presented the whole thing in a very detailed way, mainly in the first and last installments; he gives you an overview about the whole working of the system, so that the people can understand them in a much better way. One of the first stories to be published proclaimed that the rates of one prenatal drug could increase 16 fold and this would be the result of the approval by the FDA. Due to this, the local pharmaceutical company would end up earning tons of money. The second story published is quite an interesting one. It concerns the criticizing of a former chief of the same company by a US District Judge. The Judge condemned him by accusing him of power abuse, greed and being reckless in his handling of the affairs of the firm.

But he concludes his reports with a stern warning. He is of the opinion that there would huge long terms costs involved because of the quick approval by the FDA. These costs could medical as well as the monetary part. So he is indirectly citing that the fast track approvals of the drugs by KV pharmaceuticals might not be a good move and that it could have some serious implications in the future. The rates of the drugs are reaching its zenith, so its time to a timely check.

Launch of First Lupus Drug, Never Before in 56 Years

With advanced inventions and technological support, FDA (Food and drug administration) has been bringing out solutions on their years of efforts made. This new lupus drug has made a grand entry in the pharmaceutical sectors, as being the one that was worked from last 56 years. With such intense study and immense efforts, the drug named Benlysta (belimumab) finally got approved as the best solution that is approved to treat lupus.

The medicine basically aids in treating patients with autoantibody-positive lupus which is also known as systemic lupus erythematosus. The drug works to the core in such patients, despite of them involved in various therapies like corticosteroids, antimalarials, non steroidal anti-inflammatory drugs and immunosuppressive medications.

In 1948, Aspirin got approved as a solution that treats lupus. While the action mechanism of the drug is concerned; Benlysta works by directly delivering into the veins through intravenous infusion. The medication also ranges as the foremost inhibitor targeting on the B-lymphocyte stimulator (BLyS) protein; this mechanism helps to control the abnormal B cells that are responsible for creating problems in lupus.

Lupus is one of the extreme, serious and fatal autoimmune diseases ruining the functions of healthy tissues. Women are the victims, and are generally observed in those in the age group of 15 to 44. When triggered by the condition, it affects various body parts including, kidneys, lungs, joints, skin, heart and even brain. The basic symptoms of lupus can be observed through swelling of joints, pain in the joints, fever, chest pains, light sensitivity, fatigue and in some cases even hair loss.

The health condition is widely observed in United States of America, in women approximately ranging from 300000 to 1.5 million. However, the risks in African American women are three times more as compared to other areas, depending on the lifestyle they follow.

But now with the invention of Benlysta, all the lupus related worries will come to an end. It is a phenomenal existing therapy as an altogether new approach to look after and manage the troubles faced by women.

There is lot of technicalities involved when it comes to using a drug, this can be made sure with the help of a complete medical practitioner.

Sextacy, A Deadly Mix of Viagra and Ecstasy on The Wave

The police department at Calgary is saying that drugs like Viagra are being sold on the streets by combining it with Ecstasy. These Anti-Impotence drugs have attracted the attention of criminals who are finding this mix very lucrative. Drugs like Viagra and Cialis are often stolen from most drugstore heists, because the market for an illegal combination of Viagra pills and ecstasy seems to be booming on an underground level.

Apart from ed drug Viagra (or Cialis) has many other health benefits (such as it improves heart performance as Viagra dilates blood capillaries)

The Sgt who is in charge of such cases says that “ People are looking out to mix any kind of drug in order to get a high, so it wont be surprise if they make a combo of ecstasy and pills like Viagra to stimulate themselves”. Some people are labeling this mix as ‘sextacy”. Ecstasy is a very potent and illegal drug used at parties and it is known to give you a great high and Viagra gives you that good staying power.

This is the reason why people are mixing both these drugs to attain a heightened feeling. The police have said that Anti-impotence drugs are very powerful and a person using it illegally can harm his own health on a dangerous level. Such drugs should be taken only on the prescription of a doctor.

Office Hanson says “This problem of the trafficking of the pharmaceutical drugs is quite a concern for us because it can be dangerous for the user in many ways. Though I am yet to come across such cases, but the demand for such drugs in the illegal trade is pretty much high and the people will buy any drug to enhance their senses.”

Female Sexual Desire Booster Drug To be Dicussed by FDA

Reproductive Health Drugs Advisory Committee of FDA is due to discuss the request made by Boehringer Ingelheim (A giant german pharmaceutical) to approve Flibanserin to increase libido in women. In pharmaceutical inductry the drug is known as female Viagra. Flibanserin, whose proposed names are Ectris and Girosa, may be the first of its kind medicine that stimulates and increases sex desire in females.

Desire of sex in women is outcome of more complex events than that of men. Apperently, Flibanserin won't work like viagra and it won't increase blood flow in female genital region. Flibanserin works by altering the chain of chemical reactions in the brain that finally result in increased sexual desire.

Unlike other proposed treatment such Intrinsa that releases testosterone through the skin into the blood, Flibanserin manipulates events of several reactions and that is why it is believed to be more effective.
Amy Allina, a member of National Women's Health Network, advocates the importance of such drug saying that sex life can be a problematic issue for some women in order to sustain healthy sex life and thereby happy marriage life.

Market of female viagra drug is estimated to be billions of dollars as its been more than 10 years since viagra was launched and no similar drug for females since then.

Lung Cancer is Linked to Defetive ALK Gene

Use of living matters in medicines is believed to be new way to treat cancers. ALK inhibitors (a type of gene inhibitors) are likely to be new class of drug alongside others such as Monoclonal antibodies and molecule inhibitors.

Non small-cell lung cancer or NSCLC is found in 8 to 9 out of 10 cases of all lung cancers. Defective rearrangment of ALK (anaplastic lymphoma kinase) is found to be triggering event to cause NSCLC. A study conducted at University of Colorado and represented at joint conference of AARC and IASLC says that normal rearrangements of chromosomes interrupt the ALK gene causing its fusion with another gene which triggers creation of oncogenic ALK fusion genes. This event initiates cell growth.

A drug, named Crizotinib in experiments, by Pfizer has shown positive result in lung tumers. Experiments showed reduction of tumor by 57% and progression of tumor stopped in 87% of the patients.

Crizotinib is said to be only drug that is intended to target defective gene. The drug could pave the way for new therapeutic mechanism in drugs used for cancer.

Alice Shaw, a thoracic cancer specialist at Massachusetts General Hospital in Boston explains how rearrangements turns into cancer: Normal chromosomal arrangement of defective ALK gene breaks and part of broken gene gets fused with other gene. Such rearrangements cause growth of cells out of control.
Crizotinib is under experiments and could hit the market in 2013 if found positive results in later large scale studies.

Nasal Spray To Relieve Pain

Recently FDA has approved Sprix (with ketorolac tromethamine as active ingredient) to treat acute pain. Proposed use of Sprix is to relieve kind of pain that is required to treat without any opiod pain killer medications. It comes as better option to avoid opiod medicines which are highly addicative in nature. Sprix is NSAIDs, ketorolac is a non-selective COX inhibitor.

Current use of Ketorolac as injectable medicine to manage pain is widely accepted by physicians in hospitals. Being in a convenient form (nasal spray) Sprix is expected to be popular as instant pain killer remedy. Because of the way of uptake Sprix shows fast effects. It is absorbed fast in blood and acts as rapidly as to its injection counterpart.

Being rapid in action and fast effects, Sprix is classified as prescription medication. Physicians warn against its use in terms of self-medication. Even little overdose may cause serious side effects. Its use is strictly limited to 5 days.

Nasal pain killer spray is designed to provide ambulatory patients a convenient, highly effective, and rapid action alternative to treat acute moderate to moderately severe pain. Ketorolac tromethamine is a currently administered in USA hospitals as non-narcotic injectable pain killer for moderately severe pain.
